Solution
No, Using both PCI-Express interfaces simultaneously is prohibited.
Notes:
The board should be removed from your PC when the PCI-Express cable connection is used.
The board should be disconnected from the PCI-Express cable when PCI-Express edge connector is used (inside PC)
